Discovery of a Novel Inhibitor of Human Purine Nucleoside Phosphorylase by a Simple Hydrophilic Interaction Liquid Chromatography Enzymatic Assay

Summary: This study presents the discovery of novel HsPNP inhibitors through the combination of experimental and computational tools, developing a simple, inexpensive, and non-radioactive enzymatic assay for screening inhibitors, and evaluating the cytotoxicity of inhibitors on T-cell leukemia cells. The inhibition potency of the most promising compound was quantified, and docking studies provided insights into the inhibitor interaction at the HsPNP active site, offering new tools and leads for developing novel HsPNP inhibitors.
