Super-resolved 3D tracking of cargo transport through nuclear pore complexes

Summary: The study introduces a multi-color imaging approach that allows for the visualization of cargo transport trajectories relative to a super-resolved nuclear pore complex scaffold in non-fixed permeabilized cells. The findings demonstrate the presence of hourglass-shaped translocation conduits near the NPC scaffold, through which cargo complexes can rapidly migrate across the permeability barrier. The binding sites for cargo complexes extend beyond 100 nm from the pore openings.
