Three-dimensional interactions between enhancers and promoters during intestinal differentiation depend upon HNF4

Lei Chen et al.

Summary: In renewing tissues like the intestinal epithelium, enhancer-promoter interactions remain relatively stable despite dynamic gene regulation across the differentiation axis. HNF4 transcription factors are found to be essential for chromatin looping at target genes, with depletion of HNF4 disrupting local chromatin looping, histone modifications, and target gene expression. This study offers insights into the transcriptional regulatory mechanisms governing homeostasis in renewing tissues.

The nuclear receptor HNF4 drives a brush border gene program conserved across murine intestine, kidney, and embryonic yolk sac

Lei Chen et al.

Summary: The brush border is a specialized structure on the apical surface of epithelia that greatly increases absorptive surface area, but the transcriptional regulatory networks controlling brush border genes are not fully understood. This study identifies HNF4 as a conserved regulator of the brush border gene program in multiple organs, such as intestine, kidney, and yolk sac, and reveals its role in chromatin looping between promoters and enhancers in the intestine.

Analysis of 1,25-Dihydroxyvitamin D3 Genomic Action Reveals Calcium-Regulating and Calcium-Independent Effects in Mouse Intestine and Human Enteroids

Shanshan Li et al.

Summary: Transgenic expression of vitamin D receptor (VDR) in the distal intestine is crucial for normalizing serum calcium levels and rescuing rickets in VDR null mice. The activation of certain genes in the proximal and distal intestine by 1,25(OH)(2)D-3 indicates an interrelationship between vitamin D and intestinal calcium transport. Additionally, the conservation of the effects of 1,25(OH)(2)D-3 in mice and humans suggests a significant role of distal intestinal segments in vitamin D action.
