Highly sensitive and specific detection of hepatitis B virus DNA and drug resistance mutations utilizing the PCR-based CRISPR-Cas13a system

S. Wang et al.

Summary: A highly sensitive and practical method for HBV and drug resistance detection using PCR-CRISPR was established, with the ability to detect one copy per test of HBV DNA and YMDD drug resistance mutations. The method has wide application prospects for early detection of HBV infection, drug resistance monitoring, and treatment guidance.

Emergence and dissemination of three mild outbreaks of Neisseria gonorrhoeae with high-level resistance to azithromycin in Barcelona, 2016-18

P. Salmeron et al.

Summary: This study characterized the first 16 Neisseria gonorrhoeae isolates with high-level azithromycin resistance (HL-AziR) in Barcelona between 2016 and 2018. WGS was used to identify antimicrobial resistance mechanisms and clonal relatedness, revealing three transmission clusters associated with sexual behavior. The findings highlight the continuous development and spread of antimicrobial resistance in N. gonorrhoeae, supporting enhanced outbreak investigation and public health measures for prevention and control of multi-drug resistant strains.

Sustained Transmission of Neisseria gonorrhoeae with High-Level Resistance to Azithromycin, in Indianapolis, Indiana, 2017-2018

Justin L. Holderman et al.

Summary: A cluster of 14 Neisseria gonorrhoeae isolates with high-level azithromycin resistance was identified in Indiana, primarily among white men who have sex with men. Although few patients named each other as partners, genomic sequencing revealed isolates as closely related.

The Mosaic mtr Locus as Major Genetic Determinant of Azithromycin Resistance of Neisseria gonorrhoeae-Germany, 2018

Sebastian Banhart et al.

Summary: The resistance of Neisseria gonorrhoeae to azithromycin within the German Gonococcal Resistance Network increased from 4.3% in 2016 to 9.2% in 2018. The rise in resistance was primarily clonal and linked to a specific mtr locus. The international spread of resistant lineages was identified as a major factor contributing to azithromycin resistance in Germany.

Molecular pathways to high-level azithromycin resistance in Neisseria gonorrhoeae

J. G. E. Laumen et al.

Summary: This study characterized the genetic pathways leading to high-level azithromycin resistance in Neisseria gonorrhoeae by using a customized morbidostat to subject reference strains to sustained drug pressure. The results showed stepwise evolution of resistance, with key mutations identified in ribosomal proteins and efflux pumps. High-level resistance was consistently associated with mutations in the 23S ribosomal RNA, including well-known A2059G and C2611T mutations.
