Innate immunity drives pathogenesis of rheumatoid arthritis

Maria Edilova et al.

Summary: Rheumatoid arthritis is an autoimmune disease affecting about 1% of the population, with genetic and environmental factors playing key roles in its pathogenesis. While early studies focused on lymphocytes, recent research has highlighted the critical involvement of innate immune cells in the onset and progression of RA. Therapeutic approaches targeting different inflammatory pathways and immune cells, including the emerging roles of innate lymphoid cells and inflammasomes, are being explored for the treatment of RA.

In vitro elimination of autoreactive B cells from rheumatoid arthritis patients by universal chimeric antigen receptor T cells

Bo Zhang et al.

Summary: Autoreactive B cells play a crucial role in the pathogenesis of rheumatoid arthritis (RA), and targeting these cells using FITC-labelled antigenic peptide epitopes with universal CAR-T cells shows promise for precise and customized treatment of RA and potentially other systemic autoimmune diseases.

Impact of Posttranslational Modification in Pathogenesis of Rheumatoid Arthritis: Focusing on Citrullination, Carbamylation, and Acetylation

Eui-Jong Kwon et al.

Summary: Rheumatoid arthritis (RA) is triggered by a complex interplay of genetic, environmental, and immunologic factors, with posttranslational modifications and cell death mechanisms playing a role in the disease's pathogenesis. Certain microbial infections may induce autoantigens, while anti-modified protein antibodies are crucial in prediction, diagnosis, and prognosis of RA. Smoking is associated with both PTMs and AMPAs in RA development, although evidence linking smoking to the generation of AMPAs is currently lacking.

B Cells in Rheumatoid Arthritis:Pathogenic Mechanisms and Treatment Prospects

Fengping Wu et al.

Summary: Rheumatoid arthritis is a common, chronic, systemic autoimmune disease characterized by immune system disorders, with autoreactive CD4(+)T cells, pathogenic B cells, and inflammatory cytokines being key players. Further research is needed to distinguish between pathogenic B cells and protective B cells in patients with RA to develop more targeted treatments.

Function and Role of Regulatory T Cells in Rheumatoid Arthritis

Qi Jiang et al.

Summary: Rheumatoid arthritis is a systemic and heterogeneous autoimmune disease characterized by symmetrical polyarthritis, with dysfunction of regulatory T (Treg) cells potentially contributing to the breakdown of self-tolerance. The ideal treatment strategy for RA should focus on re-inducing self-tolerance to prevent obvious tissue injury.

Autoantibodies in Rheumatoid Arthritis - Laboratory and Clinical Perspectives

Johan Ronnelid et al.

Summary: Measurement of autoantibodies like RF and ACPA is crucial in the diagnosis and treatment of RA, but lack of harmonization between different commercial tests poses a challenge. It is recommended that all RF and ACPA tests should have a diagnostic specificity of 98-99%, and the introduction of test result-specific likelihood ratios can provide more precise information for clinical decision-making. Detection of autoantibodies can also be used to predict future arthritis development and prognosis in at-risk patients.

Summary: Rheumatoid arthritis is influenced by genetic, environmental, and immunological factors, with protein posttranslational modifications, cell death mechanisms, and autoantigenicity playing crucial roles in its pathogenesis. Microbial infections can induce autoantigens, while anti-modified protein antibodies are important for prediction, diagnosis, and prognosis. Smoking is associated with both posttranslational modifications and anti-modified protein antibodies in RA, although evidence of smoking inducing their generation is lacking.
