Summary: High-risk neuroblastoma, often associated with MYCN oncogene amplification, may benefit from treatment with ALK tyrosine kinase inhibitors targeting activating ALK mutations. Overexpression of ALKAL2 ligand in NB may exacerbate the disease progression, even in the absence of ALK mutations.

Mapping the Phospho-dependent ALK Interactome to Identify Novel Components in ALK Signaling

Summary: This study systematically characterized the phospho-dependent ALK interactome using the MaMTH system, identifying 30 novel ALK interactors from a library of 86 SH2 domain-containing full length proteins. The interactions of these novel proteins are correlated to ALK phosphorylation activity and NCK2 was further validated as a key interactor in neuroblastoma cells. The study provides a valuable resource list of potential novel ALK signaling components for further exploration.

Summary: The development of sympathetic trunks in human embryos was visualized using 3D-reconstruction software, revealing a change in position from lateral to dorsolateral as a criterion to distinguish primary and secondary trunks. The differential growth accounted for the regional differences in position of the sympathetic trunks as they transitioned from para-aortic to prevertebral or paravertebral positions during development.
