Bioinspired Thermoresponsive Xyloglucan-Cellulose Nanocrystal Hydrogels

Malika Talantikite et al.

Summary: This study reported the preparation and application of thermoresponsive hydrogels composed of xyloglucan and cellulose nanocrystals, showing that the properties of the gels could be adjusted by specific mixing ratios and exhibiting a reversible thermal transition behavior. This thermoresponsive behavior provides opportunities for wide applications, especially in the biomedical field, where the mechanical properties could be further tuned by adjusting the content of cellulose nanocrystals.

Cellulose-hemicellulose interactions-A nanoscale view

Ali Khodayari et al.

Summary: In this work, the interactions of five different hemicellulose models with cellulose nanocrystals were studied through molecular dynamics simulations. It was found that glucuronoarabinoxylan had the highest binding free energy, while xyloglucan showed the lowest interaction energies. The presence of water molecules was observed to weaken the interactions between hemicellulose and cellulose.

Cell wall modification by the xyloglucan endotransglucosylase/hydrolase XTH19 influences freezing tolerance after cold and sub-zero acclimation

Daisuke Takahashi et al.

Summary: This study characterized the cell walls of Arabidopsis thaliana using a combination of immunohistochemical, chemical, and spectroscopic analyses, and identified a mutant in the XTH19 gene with reduced freezing tolerance after cold and sub-zero acclimation. Differences in cell wall composition and structure were associated with the reduced freezing tolerance in the mutant compared to the wild type, shedding new light on the potential roles of cell wall remodelling for the increased freezing tolerance observed after low temperature acclimation.

Entropy drives the adsorption of xyloglucan to cellulose surfaces-A molecular dynamics study

Saina Kishani et al.

Summary: This study investigated the thermodynamics of the adsorption of xyloglucan (XG) to cellulose surfaces at different temperatures using molecular dynamics simulations. The results showed that adsorption near room temperature is an endothermic process driven by the entropy of released interfacial water molecules. In the case of native cellulose, adsorption became exothermic at higher temperatures, while the interactions between water and charged groups of oxidized cellulose hindered this transition.

Enzymatic characterization of ancestral/group-IV clade xyloglucan endotransglycosylase/hydrolase enzymes reveals broad substrate specificities

Merve Seven et al.

Summary: Xyloglucan endotransglycosylase/hydrolase (XTH) enzymes have been found to exhibit broad substrate specificities, including cellulose analogues and mixed-link beta-glucans, in addition to the standard xyloglucan. This discovery opens up a wide range of potential roles for these enzymes, both within plants and in various industries. Furthermore, genome screening and expression analyses suggest that the XTH genes in the "ancestral" clades are unique to angiosperms, predominantly expressed in reproductive tissues, leading to the proposal of renaming this phylogenetic group as the group-IV clade.

Ride to cell wall: Arabidopsis XTH11, XTH29 and XTH33 exhibit different secretion pathways and responses to heat and drought stress

Monica De Caroli et al.

Summary: The study compares the secretion patterns of three members of the Arabidopsis thaliana XTH family and reveals that XTH11 and XTH33 use a conventional protein secretion pathway to reach the cell wall and plasma membrane, while XTH29 follows an unconventional protein secretion pathway towards the apoplast. The presence of a 45-amino acid extraterminal region is crucial for the correct cell wall targeting of XTH29. Furthermore, XTH29 is significantly upregulated under drought and heat stress compared to XTH11 and XTH33, indicating different involvement of these XTHs in cell wall remodeling under abiotic stress.

Carboxylated-xyloglucan and peptide amphiphile co-assembly in wound healing

Summary: This work presents a self-assembling hydrogel dressing that mimics the hierarchical structure of skin extracellular matrix by investigating the co-assembly behavior of a carboxylated variant of xyloglucan (CXG) with a peptide amphiphile (PA-H3) to generate hierarchical constructs with tuneable molecular composition, structure, and properties. The hydrogel dressing is characterized by its morphology and mechanical properties, and a preliminary biological evaluation has been carried out both in vitro with HaCat cells and in vivo in a mouse model.
