Sigma-1 receptor activity in primary sensory neurons is a critical driver of neuropathic pain

Seung Min Shin et al.

Summary: This study found that the Sigma-1 receptor is highly expressed in the primary sensory neurons involved in pain. By using gene interference, the researchers successfully targeted and reduced the expression of Sigma-1 receptor, leading to a decrease in mechanical and thermal hypersensitivity associated with neuropathic pain. These results suggest that the Sigma-1 receptor plays a critical role in regulating pain pathways in sensory neurons.

Development of New Benzylpiperazine Derivatives as σ1 Receptor Ligands with in Vivo Antinociceptive and Anti-Allodynic Effects

Giuseppe Romeo et al.

Summary: The study focused on designing new benzylpiperazine derivatives with high affinity for sigma R-1 receptors and improved selectivity over sigma R-2 receptors. The compound 15 demonstrated potent antinociceptive effects in mouse models of inflammatory pain and neuropathic pain, without causing sedation or impairing locomotor responses, showing promise as a potential therapeutic for chronic pain.

Combination of Heme Oxygenase-1 Inhibition and Sigma Receptor Modulation for Anticancer Activity

Giuseppe Romeo et al.

Summary: Targeting both HO-1 and sigma R proteins simultaneously may provide an effective strategy for treating prostate and brain cancers, leading to increased antiproliferative activity. By synthesizing novel hybrid compounds, further optimization of structure can be achieved to develop more potent anticancer agents.
