Summary: Bromodomain and extraterminal (BET) proteins are potential therapeutic targets for hematological and solid tumors, but BET inhibitor monotherapy has limited benefits for hepatocellular carcinoma (HCC). This study reveals that YAP/TAZ genes are key in determining sensitivity to BET inhibitors, and the combination of tadalafil and JQ-1 can effectively suppress HCC progression by targeting the PDE5/PKG/Hippo/YAP/TAZ axis to overcome BET inhibitor resistance.
