Predictive biomarkers for 5-ALA-PDT can lead to personalized treatments and overcome tumor-specific resistances

Maria Mastrangelopoulou et al.

Summary: This study aims to explore potential biomarkers that can predict treatment outcomes of 5-ALA-PDT and identify methods to overcome cancer cells' resistance to the therapy. The results show that the expression levels of ABCG2 transporters, FECH enzyme, and HO-1 enzyme can serve as predictive biomarkers for 5-ALA-PDT. In addition, inhibiting specific molecular targets can significantly enhance the efficacy of 5-ALA-PDT for different types of cancer cells.

Increased DNA repair capacity augments resistance of glioblastoma cells to photodynamic therapy

Somayeh Shahmoradi Ghahe et al.

Summary: Resistance to photodynamic therapy in cancer cells may be caused by various mechanisms, including efficient repair of oxidative DNA damage and DNA breaks. Higher activity of APE1 endonuclease and increased expression and activation of DNA damage kinase ATM in the U-87 MGR cell line suggest that they are potential targets for sensitizing resistant cells to PDT.

Summary: A novel approach of using a nonplanar near-infrared absorbing hyperthermia and superoxide radical photogenerator TB against hypoxic tumors was proposed. The encapsulation of TB in phototheranostic nanoparticles (TB NPs) enhanced its stability and effectiveness, demonstrating satisfactory therapeutic efficacy in eradicating hypoxic tumors.
