The emerging role of non-coding RNAs in the epigenetic regulation of pediatric cancers

Anup S. Pathania et al.

Summary: Epigenetics is a process that regulates gene expression without altering DNA sequence. Numerous studies have shown that epigenetic mechanisms play a critical role in cell growth, differentiation, and cancer. Non-coding RNAs derived from junk DNA are involved in cancer initiation, progression, and therapy resistance. However, our current understanding of their biological functions, especially in cancers, is inadequate. Understanding RNA epigenetics can help design novel therapeutics targeting epigenetic regulatory networks for the benefit of cancer patients.

Piwi-interacting RNAs (piRNAs) as potential biomarkers and therapeutic targets for cardiovascular diseases

Min Li et al.

Summary: Piwi-interacting RNAs (piRNAs) are a novel cluster of small non-coding RNAs that have been found to be widely expressed in human tissues and potentially regulate various diseases, including cardiovascular diseases (CVDs). However, the molecular mechanisms and signaling pathways involved with piRNA function have not been fully elucidated. Further research is needed to explore the potential applications of piRNAs in the clinical diagnosis, prognosis, and therapeutic strategies for heart diseases.

Population genomics in the arboviral vector Aedes aegypti reveals the genomic architecture and evolution of endogenous viral elements

Summary: Horizontal gene transfer from viruses to eukaryotic cells is a common phenomenon, with nrEVEs in Aedes aegypti potentially serving as a heritable, sequence-specific mechanism for antiviral immunity. The study shows that virus endogenization events are complex, with only a few nrEVEs playing a role in adaptive evolution in A. aegypti.

Comprehensive annotation and characterization of planarian tRNA and tRNA-derived fragments (tRFs)

Vairavan Lakshmanan et al.

Summary: tRNA-derived fragments (tRFs) are dynamically expressed during planarian regeneration, suggesting a possible role in regulating regeneration processes. Spatial profiling reveals diverse expression patterns of tRFs, with some enriched in specific regions. 5'-tsRNA interacts with SMEDWI proteins and AGO1 plays a role in processing itRFs, indicating a novel regulatory mechanism in planarian regeneration.

piRNA-mediated gene regulation and adaptation to sex-specific transposon expression in D. melanogaster male germline

Summary: Small noncoding piRNAs guide repression of complementary targets in Metazoa. Previous studies in fruit fly ovaries have shown the function of the piRNA pathway in transposon silencing and genome defense. In fruit fly testes, piRNA programs exhibit sexual dimorphism and are used differently between sexes to combat sex-specific transposon landscapes.

Establishment of H3K9me3-dependent heterochromatin during embryogenesis in Drosophila miranda

Kevin H-C Wei et al.

Summary: The study reveals that heterochromatin in Drosophila embryos is established through H3K9me3 enrichment over repetitive sequences, with initial nucleation sites appearing in specific retrotransposon families and gradually broadening into stable domains. Early nucleating transposable elements are associated with maternal piRNAs and early zygotic transcription, suggesting a model of piRNA-associated co-transcriptional silencing and additional mechanisms for site-restricted H3K9me3 nucleation at TEs in pre-cellular Drosophila embryos.
