Mechanical stress influences the morphology and function of human uterosacral ligament fibroblasts and activates the p38 MAPK pathway

Yapei Zhu et al.

Summary: The study found that hUSLFs from POP patients were larger and longer with rearrangement of F-actin. Collagen I and III expression were downregulated, MMP1 expression was upregulated, and apoptosis rate increased in POP patients. Mechanical stress induced cell morphological changes, ECM remodeling, and apoptosis, with potential involvement of the p38 MAPK pathway.

Effect of zymosan on the expression and function of the gap-junction protein connexin 43 in human cornea fibroblasts

Xiao-Shuo Zheng et al.

Summary: The ligand zymosan found on the surface of fungi has been found to reduce connexin 43 (Cx43) protein and mRNA levels as well as significantly inhibit gap junctional intercellular communication (GJIC) activity in cultured human corneal fibroblasts (HCFs). These effects were attenuated by inhibitors of the NF-kappa B, MAPK/ERK, and JNK signaling pathways, suggesting that zymosan-induced damage to corneal stroma during fungal infection may be mediated through these pathways.

Responsiveness of human bronchial fibroblasts and epithelial cells from asthmatic and non-asthmatic donors to the transforming growth factor-β1 in epithelial-mesenchymal trophic unit model

Milena Paw et al.

Summary: Asthmatic bronchial epithelial cells and fibroblasts show higher sensitivity to TGF-beta(1) and have a greater potential for fibrotic responses, with a noticeable impact on fibroblast-to-myofibroblast transition. Fibroblasts also have protective effects on TGF-beta(1)-exposed bronchial epithelial cells, affecting the proper condition and function of the EMTU in both normal and asthmatic individuals.

JNK and p38 Inhibitors Prevent Transforming Growth Factor-β1-Induced Myofibroblast Transdifferentiation in Human Graves' Orbital Fibroblasts

Tzu-Yu Hou et al.

Summary: This study found that TGF-beta 1 induced myofibroblast transdifferentiation in human Graves' orbital fibroblasts through the p38 and JNK pathways, resulting in upregulation of CTGF, α-SMA, fibronectin, TIMP-1, and TIMP-3, while inhibiting the activities of MMP-2/-9.

Increased myofibroblasts in the small airways, and relationship to remodelling and functional changes in smokers and COPD patients: potential role of epithelial-mesenchymal transition

Mathew Suji Eapen et al.

Summary: Small airway myofibroblasts play a role in airway remodelling and pathology in smokers and COPD patients, with increased presence of alpha SMA(+) cells in subepithelial Rbm, lamina propria and adventitia. The thickening of subepithelial lamina propria and adventitia, along with the increase in myofibroblasts in the lamina propria, are strongly associated with lung function decline, ECM protein deposition, and EMT activity in epithelial cells.

Mechanotransduction via the coordinated actions of integrins, PI3K signaling and Connexin hemichannels

Manuel A. Riquelme et al.

Summary: Mechanical loading activates integrin alpha 5 and alpha V, leading to the opening of connexin 43 (Cx43) hemichannels (HCs) and release of bone anabolic factors. Inhibition of integrin alpha V affects HCs opening and disrupts PI3K/AKT signaling pathway, while integrin alpha 5 activation is dependent on PI3K/AKT activation. Dendritic alpha V is a more sensitive mechanosensor than alpha 5 in activating HCs at low fluid shear stress levels.

Decoding myofibroblast origins in human kidney fibrosis

Christoph Kuppe et al.

Summary: Using single-cell RNA sequencing, researchers profiled the transcriptomes of healthy and fibrotic human kidneys, identifying distinct subpopulations of pericytes and fibroblasts as the main sources of scar-forming myofibroblasts during kidney fibrosis. This study shed light on the cellular origins and differentiation of myofibroblasts in human kidney fibrosis, identifying potential therapeutic targets such as NKD2.

Molecular and cellular mechanisms of liver fibrosis and its regression

Tatiana Kisseleva et al.

Summary: Chronic liver injury results in liver inflammation and fibrosis, which can be reversed by eliminating activated myofibroblasts and resorbing the fibrous scar. Understanding the molecular mechanisms underlying liver fibrosis and its reversibility can lead to the identification of new therapeutic targets for liver fibrosis treatment.
