Antitumoral effects of dovitinib in triple-negative breast cancer are synergized by calcitriol in vivo and in vitro

Summary: The study found that the combination of multikinase inhibitor dovitinib and natural anticancer hormone calcitriol synergistically inhibited tumor growth in TNBC, leading to a significant reduction in the dose of dovitinib while maintaining its antiproliferative potency. The dual treatment induced cell death and DNA damage, inhibited tumor growth, and decreased angiogenesis in TNBC tumors, suggesting a beneficial convergence of independent antitumor mechanisms of dovitinib and calcitriol.

Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Summary: Both the vitamin D active metabolite and synthetic analog can regulate the growth of not only estrogen receptor-positive cells, but also hormone-independent cancer cells, while combined with An, PRI-2191 and PRI-2205 significantly inhibit the tumor growth of MCF-7 cells. The potentiation of the antitumor activity in combined treatment of MCF-7 tumor-bearing mice is related to the reduced activity of aromatase and regulation of the expression of estrogen receptor ER alpha and VDR.
