Cellular senescence in ageing: from mechanisms to therapeutic opportunities

Raffaella Di Micco et al.

Summary: Cellular senescence is characterized by permanent proliferation arrest and can occur in response to various stresses like telomere dysfunction. Apart from stress-induced senescence, it can also be part of controlled programs in biological processes like embryonic development. Senescent cells contribute to impaired tissue regeneration, chronic diseases, and organismal aging, but also present an opportunity for intervention in aging and associated diseases.

Cellular senescence and its role in white adipose tissue

Ulf Smith et al.

Summary: Cell senescence results in irreversible cell cycle arrest and the induction of a senescence-associated secretory phenotype (SASP), impacting cell proliferation and influencing the development of chronic diseases like diabetes and cardiovascular disease. Efforts to target senescent cells using senolytic compounds show promise in improving chronic disorders in experimental animal models.

Effects of Long-Term DHA Supplementation and Physical Exercise on Non-Alcoholic Fatty Liver Development in Obese Aged Female Mice

Jinchunzi Yang et al.

Summary: The study found that long-term intake of a DHA-rich diet and aerobic exercise can effectively improve NAFLD in aged obese mice. The DHA diet and exercise help reduce liver steatosis, promote lipid oxidation, decrease proinflammatory gene expression, and prevent alterations in stress-related and autophagy genes induced by a high-fat diet.

LIFR-α-dependent adipocyte signaling in obesity limits adipose expansion contributing to fatty liver disease

Summary: This study revealed the importance of obesity-associated LIFR-alpha/JAK/STAT3 inflammatory signaling in adipocytes, which can block further adipose expansion in diet-induced obesity (DIO) and contribute to ectopic liver triacylglyceride accumulation.

Changes in brown adipose tissue lipid mediator signatures with aging, obesity, and DHA supplementation in female mice

Elisa Felix-Soriano et al.

Summary: The dysfunction of brown adipose tissue (BAT) in aging and obesity may be related to a decrease in production of specialized proresolving lipid mediators (SPMs), especially under chronic unresolved inflammation. While a diet rich in docosahexaenoic acid (DHA) can partially restore BAT dysfunction, it does not significantly alter the levels of specific SPMs. Despite increasing DHA-derived SPMs, it does not improve the response of BAT to cold exposure in obese-aged mice.

Synergizing Mouse and Human Studies to Understand the Heterogeneity of Obesity

Penny Gordon-Larsen et al.

Summary: This review discusses the fundamental gaps in knowledge of obesity heterogeneity and advocates for approaches to overcome limitations in the field. Best scientific practices are suggested to ensure strong rigor for investigators to delineate the sources of heterogeneity in the population with obesity. It is proposed that obesity should be considered as a heterogeneous disease with complex mechanisms and etiologies, requiring unique prevention and treatment strategies tailored to the individual.

The good and the BAT of metabolic sex differences in thermogenic human adipose tissue

Michaela Keuper et al.

Summary: This review discusses the studies on human sex differences in thermogenic adipose tissue and its impact on systemic energy metabolism. Women appear to have a higher prevalence of thermogenic adipose tissue, but the effects on metabolic disease progression and mechanisms are still unclear. Further research on sex-specific adipose metabolism in experimental animal studies may help establish molecular mechanisms for future investigations in humans.

The AMPK/p27Kip1 Pathway as a Novel Target to Promote Autophagy and Resilience in Aged Cells

Lauren K. McKay et al.

Summary: p27(Kip1) was previously thought to only act as a cyclin-dependent kinase inhibitor, but it is now recognized as a crucial mediator of various cellular processes, including autophagy, cytoskeletal dynamics, cell migration, and apoptosis. The subcellular location of p27(Kip1) plays a critical role in determining its function, with cytoplasmic p27(Kip1) promoting cellular resilience and nuclear p27(Kip1) inhibiting cell cycle progression. Regulation of p27(Kip1) cellular localization through phosphorylation by kinases like Akt and AMPK contributes to its role in aging and cell fate.
