Protein kinase inhibitors for amyotrophic lateral sclerosis therapy

Valle Palomo et al.

Summary: ALS is a fatal neurodegenerative disorder with no effective treatment currently available. Multiple pathological mechanisms such as abnormal protein aggregation, neuroinflammation and dysregulation of the ubiquitin proteasome system are involved in the neuropathology of this disease. The lack of reliable animal models has hindered drug development for ALS.

C9orf72 ALS-FTD: recent evidence for dysregulation of the autophagy-lysosome pathway at multiple levels

Jimmy Beckers et al.

Summary: Amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD) are two distinct classes of neurodegenerative disorders, sharing genetic, cellular, and molecular features. Common features include hexanucleotide repeat expansions in the C9orf72 gene and accumulation of toxic protein aggregates in affected individuals' nervous systems, potentially caused by toxic gain of function from transcribed HRE RNA or DPRs. Furthermore, alterations in protein homeostasis are suggested as a root cause of disease pathogenesis.

Molecular Mechanisms Underlying TDP-43 Pathology in Cellular and Animal Models of ALS and FTLD

Alistair Wood et al.

Summary: ALS and FTLD are neurodegenerative disorders with pathological, clinical, and genetic overlaps. The primary pathological protein, TDP-43, is observed in aggregates in affected tissues in majority of cases. Disease pathogenesis involves changes in RNA splicing, abnormal stress granules, mitochondrial dysfunction, and other cellular processes.

A RIPK1-regulated inflammatory microglial state in amyotrophic lateral sclerosis

Lauren Mifflin et al.

Summary: The study identifies a subclass of microglia, termed RIPK1-Regulated Inflammatory Microglia (RRIMs), in mouse models of ALS. RRIMs show significant up-regulation of proinflammatory pathways, are highly regulated by TNF alpha signaling, and their prevalence can be suppressed by inhibiting RIPK1 activity.

Distinct responses of human peripheral blood cells to different misfolded protein oligomers

Magdalena Leal-Lasarte et al.

Summary: Recent studies show that different types of protein misfolded oligomers have varying effects on the peripheral immune system, with some resembling responses seen in Parkinson's disease and others in Alzheimer's disease. Further research into the impact of protein misfolded oligomers on the immune system could lead to the development of blood-based diagnostic methods.

Lysosome dysfunction as a cause of neurodegenerative diseases: Lessons from frontotemporal dementia and amyotrophic lateral sclerosis

Jessica Root et al.

Summary: Frontotemporal dementia (FTD) and amyotrophic lateral sclerosis (ALS) are fatal neurodegenerative disorders linked by shared genetic causes, and lysosomal dysfunction is identified as an important pathological mechanism in both FTD and ALS, caused by genetic mutations or toxic-gain of function. Mutations in genes important for endosome-lysosome function are also found in other agedependent neurodegenerative diseases, indicating a potential commonality in lysosomal dysfunction across different diseases.

Translational evidence for lithium-induced brain plasticity and neuroprotection in the treatment of neuropsychiatric disorders

Stefano Puglisi-Allegra et al.

Summary: Increasing evidence suggests the efficacy of lithium (Li+) in neuropsychiatry, indicating common dysfunctional molecular and morphological mechanisms among psychiatric and neurological disorders. The plasticity in distinct brain circuitries also overlaps in these disorders, highlighting the potential for a multi-drug strategy to target shared features.
