The potential involvement of P2X7 receptor in COVID-19 pathogenesis: A new therapeutic target?

Paulo A. F. Pacheco et al.

Summary: Research suggests that the pathogenesis of COVID-19 may involve a hyperinflammatory response, with the activation of cytokines such as IL-1β and the NLRP3 inflammasome. The recognition of extracellular ATP by the P2X7 receptor may be related to immune dysregulation caused by the SARS-CoV-2 infection.

Purinergic Signaling of ATP in COVID-19 Associated Guillain-Barre Syndrome

Julia Leao Batista Simoes et al.

Summary: COVID-19, declared a global public health emergency, has been shown to affect not only the respiratory system, but also the nervous system, with potential links to Guillain-Barre syndrome. In addition to causing widespread inflammation, the novel coronavirus also has neurotropism, leading to neurological sequelae and increased mortality risk. Therefore, modulation of purinergic signaling and inhibition of P2X7R may be potential therapeutic approaches, and alternative measures must be considered to address the severity of critical cases related to COVID-19.

Diabetes and hypertension: Pivotal involvement of purinergic signaling

Karine Paula Reichert et al.

Summary: This review focuses on the role of the purinergic system in diabetes and hypertension, highlighting changes in the expression of P1 and P2 receptors, as well as the activities of CD39, CD73, and ADA. It suggests that manipulating the purinergic axis at various levels could potentially prevent or exacerbate the development of diabetes and hypertension as a compensatory mechanism.

Summary: SARS-CoV-2 can cause severe conditions, especially in multiple sclerosis patients, due to high levels of pro-inflammatory cytokines. Studies suggest that modulation of the cholinergic anti-inflammatory pathway may be effective in treating neuroinflammation.

COVID-19-associated coagulopathy and disseminated intravascular coagulation

Summary: The pathology of COVID-19 is exacerbated by thrombosis, DIC, and cytokine storms. D-dimer levels are commonly elevated in COVID-19, but relying solely on this marker has limitations. Treatment should be individualized based on disease severity and stage, with potential for combination therapy using heparin and nafamostat in the future. Further research is needed on fibrinolytic therapy and adsorption therapy.
