Summary: The study reveals that mHtt promotes ribosome stalling and inhibits protein synthesis in HD cells, with Fmrp being upregulated but not affecting protein synthesis or ribosome stalling. There are interactions between ribosomal proteins, translating ribosomes, and mHtt, leading to a widespread shift in ribosome occupancy and unique single-codon pauses on selected mRNA targets in HD cells. This mechanistic defect of ribosomal translocation during translation elongation can be exploited for HD therapeutics.
