Peripheral changes of endocannabinoid system components in episodic and chronic migraine patients: A pilot study

Rosaria Greco et al.

Summary: The study found transcriptional changes in endocannabinoid system components in migraineurs, suggesting a role for them in migraine. These changes were detected peripherally, providing the possibility for further investigation of their role in the clinical field.

Distinct Activity of Endocannabinoid-Hydrolyzing Enzymes MAGL and FAAH in Key Regions of Peripheral and Central Nervous System Implicated in Migraine

Adriana Della Pietra et al.

Summary: This study suggests using the endogenous cannabinoid system to alleviate migraine pain by inhibiting enzymes degrading endocannabinoids for analgesic effects. The activity of MAGL was significantly higher than FAAH in trigeminal ganglia, suggesting it as a priority target for blocking peripheral mechanisms of migraine pain. In the central nervous system, both MAGL and FAAH could serve as potential targets for reducing migraine-related enhanced cortical excitability and pain transmission.

The endocannabinoid system - current implications for drug development

C. J. Fowler

Summary: This review examines compounds affecting the endocannabinoid (eCB) system, particularly in the context of pain treatment. Directly acting CB receptor ligands and indirectly acting compounds targeting enzymes show varying effects for pain treatment. Dual-acting compounds targeting multiple pathways may offer new possibilities for pain management.

Characterization of the peripheral FAAH inhibitor, URB937, in animal models of acute and chronic migraine

Rosaria Greco et al.

Summary: Inhibiting FAAH to enhance anandamide signaling shows potential for treating anxiety and pain. URB937 effectively reduces trigeminal hyperalgesia in both acute and chronic migraine animal models, likely through increased CB1 receptor activation via anandamide.

Dual Inhibition of FAAH and MAGL Counteracts Migraine-like Pain and Behavior in an Animal Model of Migraine

Rosaria Greco et al.

Summary: The study found that dual inhibition significantly reduced migraine-like features induced by nitroglycerin, possibly through cannabinoid 1 receptor-mediated action. This effect was associated with a reduction in gene expression levels of CGRP and cytokines in central and peripheral structures.
