Synthetic Protein Scaffolds for Improving R-(-)-Linalool Production in Escherichia coli

Jing Wu et al.

Summary: By physically colocalizing three enzymes in synthetic complexes, the production efficiency of R-(-)-linalool in Escherichia coli was successfully improved. Through further optimization of inducer, temperature, and glycerol concentration, the production titer in the shake flask increased by approximately 10 times compared to the scaffold-free control, with a significant increase in production yield in the fermenter as well.

Orientin and vitexin production by a one-pot enzymatic cascade of a glycosyltransferase and sucrose synthase

Cong Qiu et al.

Summary: A highly efficient biocatalytic cascade for orientin and vitexin production was developed in this study, utilizing Trollius chinensis C-glycosyltransferase (TcCGT) and Glycine max sucrose synthase (GmSUS). By modulating the catalytic efficiencies of enzymes, high titers of orientin and vitexin were achieved, with maximal titers reaching 7090 mg/L and 5050 mg/L, respectively. The method described herein can be widely used for C-glycosylation of flavonoids.

Plasticity engineering of plant monoterpene synthases and application for microbial production of monoterpenoids

Dengwei Lei et al.

Summary: Plant monoterpenoids with structural diversities have extensive applications, but their agricultural production is less effective due to geographical and seasonal dependence. Synthetic biology enables engineered microbes for efficient production, and plasticity engineering of plant MTPSs plays a key role in achieving this goal.

Production of trans-cinnamic acid by whole-cell bioconversion from l-phenylalanine in engineered Corynebacterium glutamicum

Jaewoo Son et al.

Summary: This study successfully engineered Corynebacterium glutamicum as a whole-cell catalyst for the bioconversion of l-phenylalanine into trans-cinnamic acid (t-CA) with high efficiency, reaching a production yield of 75% through repetitive supplementation. The developed bioconversion process and micromembrane-based cell recycling system provide a promising platform for industrial-scale production of t-CA using microorganisms.
