Exome or genome sequencing has identified a variety of genetic mutations associated with this syndrome, with some families receiving a presumptive molecular diagnosis. However, the majority of diagnoses are for emerging clinical conditions with characteristics that overlap the DubS phenotype.

Summary: This study describes a consanguineous Iraqi family with biallelic loss-of-function of the SLC13A5 gene, leading to a specific developmental epileptic encephalopathy with hypoplastic amelogenesis imperfecta. Analysis of the genomic architecture revealed evidence of genomic instability caused by Alu/Alu-mediated rearrangement at the SLC13A5 locus. This case highlights the importance of read-depth-based CNV detection in comprehensive exome sequencing analysis.

The structure, function and evolution of a complete human chromosome 8

Summary: The study completed the linear assembly of human chromosome 8 using long-read sequencing technologies, resolving five long-standing gaps, including regions important for disease risk. The research revealed that the centromeric alpha-satellite sequence is generally methylated, and conducted comparative analysis of centromeres in chimpanzee, orangutan and macaque. The study estimates that the mutation rate of centromeric satellite DNA is accelerated compared to unique portions of the genome.

Early consideration of LPI is important, especially in cases of early-onset systemic lupus erythematosus (SLE) with poor response to immunosuppressive therapy.
