The central role of glutathione peroxidase 4 in the regulation of ferroptosis and its implications for pro-inflammatory cytokine-mediated beta-cell death

Summary: Pro-inflammatory cytokines play a crucial role in beta-cell destruction in type 1 diabetes mellitus, while the involvement of ferroptosis in the pathogenesis of the disease has not been clearly defined. The antioxidative enzyme GPx4 is predominantly expressed in beta-cells, and its overexpression as well as the ferroptosis inhibitor ferrostatin-1 can effectively attenuate beta-cell death. Nitric oxide produced during pro-inflammatory cytokine action prevents the induction of ferroptosis, favoring apoptosis as the primary cell death mechanism in beta-cells.
