Cell cycle on the crossroad of tumorigenesis and cancer therapy

Jing Liu et al.

Summary: Cell cycle aberrancy is a fundamental mechanism underlying tumorigenesis, and therapies targeting cell cycle machinery components can not only suppress cancer cell division but also reverse cancer metabolism and restore cancer immune surveillance. Recently, proteolysis targeting chimeras (PROTACs) have been used to target oncogenic proteins related to cell cycle progression for more effective tumor suppression.

Skp2 dictates cell cycle-dependent metabolic oscillation between glycolysis and TCA cycle

Jing Liu et al.

Summary: Glucose metabolism in mammalian cells varies with different cell cycle phases, with TCA cycle being predominantly used in G1 phase and glycolysis favored in S phase. The coupling of cell cycle with metabolism is largely achieved through Skp2-mediated timely destruction of key TCA cycle enzymes IDH1/2. This mechanistic link between aberrant cell cycle and glycolysis addiction provides insights into the coupling of metabolic fluctuation with periodic cell cycle in mammalian cells.

Identification of lysine isobutyrylation as a new histone modification mark

Zhesi Zhu et al.

Summary: Lysine isobutyrylation (Kibu) is a newly discovered posttranslational chemical modification on nuclear histones, derived from isobutyryl-CoA. Various histone acetyltransferases exhibit lysine isobutyryltransferase activity in vitro, with p300 being a major regulator of histone isobutyrylation levels in cells. The X-ray crystal structures of HAT1 in complex with isobutyryl-CoA provide insight into HAT-catalyzed isobutyrylation, and RNA-Seq profiling shows that isobutyrate greatly affects gene expression in pivotal biological pathways.

The methyltransferase METTL9 mediates pervasive 1-methylhistidine modification in mammalian proteomes

Erna Davydova et al.

Summary: Post-translational methylation, specifically protein histidine methylation, is crucial for optimizing protein function. METTL9 is identified as a broad-specificity methyltransferase responsible for the majority of 1-methylhistidine modifications in mouse and human proteomes. The methylation catalyzed by METTL9 enhances respiration via Complex I, and reduces the zinc binding affinity of HxH-containing peptides.

Targeting SLC1A5 and SLC3A2/SLC7A5 as a Potential Strategy to Strengthen Anti-Tumor Immunity in the Tumor Microenvironment

Marianna Nachef et al.

Summary: Cancer cells create a hostile tumor microenvironment that challenges immune cells' metabolic fitness. Enhancing immune cell metabolism, particularly by upregulating nutrient transporters like glucose and amino acid transporters, can strengthen tumor immunotherapy efforts. Amino acid transporters not only facilitate amino acid uptake but also play a role in sensing amino acid levels and activating key metabolic pathways essential for cell growth and proliferation.

SPOP-mediated ubiquitination and degradation of PDK1 suppresses AKT kinase activity and oncogenic functions

Qiwei Jiang et al.

Summary: The study reveals that E3 ligase SPOP mediates the ubiquitination and degradation of PDK1, and mutations in SPOP impair this process, leading to activation of the AKT kinase in prostate cancer. This highlights the oncogenic roles of SPOP loss-of-function mutations and PDK1 gain-of-function mutations in tumorigenesis through AKT activation.

Choline kinase alpha 2 acts as a protein kinase to promote lipolysis of lipid droplets

Rui Liu et al.

Summary: Lipid droplets play a critical role in cancer cell growth and survival, and the protein CHKα2 is shown to be involved in promoting lipid droplet lipolysis, fatty acid oxidation, and brain tumor growth. Phosphorylation and acetylation levels of CHKα2 are positively correlated with poor prognosis in glioblastoma patients.
