Fragment Binding to Kinase Hinge: If Charge Distribution and Local pKa Shifts Mislead Popular Bioisosterism Concepts

Matthias Oebbeke et al.

Summary: In medicinal chemistry optimization, strategies involve replacing functional groups and attaching larger substituents at a promising lead scaffold. While bioisosterism rules are considered, it is challenging to determine whether the introduced modifications match the required properties at a binding site. Modulating electron density distribution and pK(a) values can influence protonation states and bioavailability. The importance lies in correctly designing pK(a) values of attached functional groups at the parent scaffold.
