Allovalency observed by transferred NOE: interactions of sulfated tyrosine residues in the N-terminal segment of CCR5 with the CCL5 chemokine

Summary: Sulfation of tyrosine residues Y14 and Y15 in the N-terminal segment of CCR5 is essential for high-affinity binding to CCL5, with disulfation resulting in up to a threefold increase in affinity. The sulfated tyrosine residues interact with the same binding pocket on CCL5 and can individually occupy the site with varying affinity, suggesting the presence of allovalency in sulfated Nt-CCR5 peptides. This allovalency, along with stronger electrostatic interactions, may contribute to the increased affinity of disulfated peptides for CCL5.
