RAD51: Beyond the break

Isabel E. Wassing et al.

Summary: RAD51, as the primary catalyst of homologous recombination (HR) in vertebrates, is not only involved in repairing double-stranded DNA breaks (DSBs), but its importance in DNA metabolism has also been increasingly recognized, with its function emerging as a double-edged sword in the interplay with DNA replication and the maintenance of genomic stability.

MYC as a target for cancer treatment

Michael J. Duffy et al.

Summary: The MYC gene, with its three paralogs, is a common driver gene in cancer, making it an attractive target for cancer treatment. While traditionally considered undruggable, recent advancements have shown potential compounds that can inhibit MYC and exhibit anticancer activity.

mTOR inhibition acts as an unexpected checkpoint in p53-mediated tumor suppression

Ning Kon et al.

Summary: Research showed that p53-4KR mice were tumor prone but failed to develop early-onset tumors; mutations at all five acetylation sites diminished p53's tumor suppression function; mTOR inhibitor can suppress early tumor formation in p53-5KR mice.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Unraveling the Molecular Tumor-Promoting Regulation of Cofilin-1 in Pancreatic Cancer

Silke D. Werle et al.

Summary: Unraveling mechanistic regulations is crucial for understanding tumor behavior and developing effective treatments. Mechanistic modeling offers a more comprehensive view of signaling pathways, identifying new therapeutic targets. In pancreatic cancer, CFL1 overexpression is associated with tumor promotion, and mechanistic modeling can predict its regulatory mechanisms and validate them through protein level analysis.

LINC00941 promotes glycolysis in pancreatic cancer by modulating the Hippo pathway

Ming Xu et al.

Summary: The study reveals that LINC00941 promotes PDAC tumor growth by enhancing aerobic glycolysis through interaction with mammalian STE20-like protein kinase 1 (MST1) and facilitating PP2A-mediated dephosphorylation of MST1, leading to activation of the Hippo pathway.

SF3B1 mutation in pancreatic cancer contributes to aerobic glycolysis and tumor growth through a PP2A-c-Myc axis

Jian-Yu Yang et al.

Summary: The SF3B1(K700E) mutation promotes tumor growth in pancreatic ductal adenocarcinoma, while knockdown of SF3B1 has a significant inhibitory effect on cell proliferation in cell lines carrying this mutation. Ectopic expression of SF3B1(K700E) restores the inhibitory role induced by SF3B1 knockdown. The mutation also promotes tumor glycolysis through the aberrant splicing of PPP2R5A and activation of c-Myc. Pharmacological activation of PP2A with FTY-720 compromises the growth advantage induced by the SF3B1(K700E) mutation.

Identification of a Small-Molecule Glucose Transporter Inhibitor, Glutipyran, That Inhibits Cancer Cell Growth

Makoto Kawatani et al.

Summary: Glutipyran and its derivatives inhibit glycolytic activity and cell growth in human pancreatic cancer cells. Proteomic profiling showed that glutipyran is clustered within endoplasmic reticulum (ER) stress inducers, including glycolysis inhibitors. Glutipyran acts as a broad-spectrum GLUT inhibitor and reduces cancer cell growth by suppressing the glycolytic and pentose phosphate pathways.
