Cathepsin B-responsive and gadolinium-labeled branched glycopolymer-PTX conjugate-derived nanotheranostics for cancer treatment

Summary: Multi-modal therapeutics using polymeric carriers for simultaneous diagnosis and treatment of cancer have been developed. A theranostic nanomedicine, branched glycopolymer-PTX-DOTA-Gd, showed extended circulation time, enhanced tumor accumulation, and excellent biocompatibility with reduced gadolinium ion retention. This branched polymeric prodrug-based nanomedicine has great potential for safe and effective cancer diagnosis and treatment, achieving enhanced imaging contrast and high antitumor efficacy.

Cold atmospheric plasma modification of curcumin loaded in tri-phosphate chitosan nanoparticles enhanced breast cancer cells apoptosis

Summary: The research aims to increase the solubility of curcumin using cold atmospheric plasma (CAP) and evaluate the effects on breast cancer cells and normal cells. By modifying curcumin through CAP and loading it into chitosan nanoparticles, the study found that the modified curcumin had inhibitory effects on MCF-7 breast cancer cells while leaving normal cells unharmed. Real-time PCR results indicated an upregulation of TP53 gene expression and downregulation of VEGF gene expression in treated MCF-7 cells.

Summary: Despite progress in cancer nanotherapeutics, obstacles like low drug loading and off-target toxicity hinder the clinical success of nanocarriers. The conjugation of docetaxel and celastrol to lactoferrin shows enhanced anti-tumor efficacy in breast cancer-bearing mice, indicating potential for synergistic therapy. This study demonstrates the proof-of-principle for coupling dual drugs to lactoferrin as a versatile nanoplatform to augment their anticancer efficacy.

In vitro experiments showed that the nanoparticles were able to down-regulate P-glycoprotein expression and improve anticancer efficacy. Additionally, NIR laser irradiation induced nanoparticles to generate reactive oxygen species, effectively inhibiting multidrug resistance and metastasis in breast cancer.
