Correlating Immune Cell Infiltration Patterns with Recurrent Somatic Mutations in Advanced Clear Cell Renal Cell Carcinoma br

Nicholas H. Chakiryan et al.

Summary: This study analyzed tumor samples from patients with metastatic kidney cancer and found that common genetic mutations are associated with distinct immune cell patterns within the tumor immune microenvironment. The study provides evidence of the interactions between cancer genetics and the immune system in kidney cancer.

BAP1-Mutated Clear Cell Renal Cell Carcinoma A Clinicopathologic Characterization of 14 Molecularly Confirmed Tumors

Alexander J. Gallan et al.

Summary: The study identifies the pathological features of BAP1-mutated CCRCC, which exhibit overlapping morphological characteristics with other subtypes of renal cell carcinoma and specific immunohistochemical patterns. BAP1-mutated CCRCCs are often aggressive, with a high likelihood of invasion and metastasis.

Meta-analysis of tumor- and T cell-intrinsic mechanisms of sensitization to checkpoint inhibition

Kevin Litchfield et al.

Summary: Checkpoint inhibitors (CPIs) enhance adaptive immunity, with clonal tumor mutation burden (TMB) identified as the strongest predictor of CPI response. Dinucleotide variants may serve as a source of immunogenic epitopes, while copy-number alterations and HLA evolutionary divergence lack pan-cancer significance.

Epigenetic silencing by SETDB1 suppresses tumour intrinsic immunogenicity

Gabriel K. Griffin et al.

Summary: A CRISPR-Cas9 screen in mouse tumour models treated with immune checkpoint blockade identified SETDB1 as an epigenetic checkpoint protein that suppresses tumour-intrinsic immunogenicity. Loss of SETDB1 leads to derepression of immune-stimulating genes and triggers TE-specific cytotoxic T cell responses, suggesting it as a potential target for immunotherapy in cancer treatment.
