Targeting Epigenetics in Lung Cancer

Yvonne L. Chao et al.

Summary: The epigenetic landscape, including DNA methylation, chromatin organization, histone modifications, and noncoding RNA regulation, plays a significant role in the heterogeneity and challenges of developing effective therapies for lung cancer. This review provides an overview of the epigenetic alterations in cancer pathogenesis and progression, as well as clinical applications for targeting epigenetics in lung cancer treatment.

A CRISPR/Cas13a-powered catalytic electrochemical biosensor for successive and highly sensitive RNA diagnostics

Yan Sheng et al.

Summary: The study introduces a dual signal amplification strategy using the CRISPR/Cas13a system and CHDC for rapid and accurate detection of low expression level RNAs in early-stage cancer. The reusable biosensor platform showed high specificity and sensitivity in distinguishing early-stage lung cancer patients from healthy subjects.

Single-cell lineages reveal the rates, routes, and drivers of metastasis in cancer xenografts

Jeffrey J. Quinn et al.

Summary: This study used a Cas9-based lineage tracer to track the development and metastasis of lung cancer cells in a mouse model over months, revealing heterogeneity in metastatic capacity and genetic differences that drive invasiveness. The study also uncovered some genes driving metastasis and an unexpected suppressive role of KRT17 in metastasis.

Sensitive Detection of KRAS Mutations by Clustered Regularly Interspaced Short Palindromic Repeats

Huifen Zhou et al.

Summary: Kirsten rat sarcoma viral oncogene (KRAS) is the most frequently mutated isoform in human tumors. This study aimed to develop CRISPR-Cas12a as a sensitive test for detecting KRAS mutations, showing a higher sensitivity compared to PCR and sequencing analysis. However, further validation in large and prospective studies is needed to determine its clinical significance.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Rlf-Mycl Gene Fusion Drives Tumorigenesis and Metastasis in a Mouse Model of Small Cell Lung Cancer

Metamia Ciampricotti et al.

Summary: A common RLF-MYCL gene fusion in human SCLC was found to accelerate tumor transformation and proliferation in a mouse model, enhancing metastatic spread. This study supports RLF-MYCL as the first validated fusion oncogenic driver in SCLC.
