Predicting Volume of Distribution in Humans: Performance of In Silico Methods for a Large Set of Structurally Diverse Clinical Compounds

Neha Murad et al.

Summary: This study advances the prediction of V-D,V-ss directly from structure with the aid of in vitro data. A large set of clinical compounds (n = 956) was rigorously evaluated using various methods. The novel data set (n = 254) generated using a single protocol for each in vitro assay presented in this study could further contribute to advancing V-D,V-ss prediction methodologies.

Summary: The study evaluated the validity of traditional mixture risk assessment paradigms IA and CA with NAMs data. Results showed that CA is more accurate in predicting mixture effects compared to IA, but in some cases, mixture bioactivity appeared to be greater than that of individual components.

Assessing Human Exposure to SVOCs in Materials, Products, and Articles: A Modular Mechanistic Framework

Clara M. A. Eichler et al.

Summary: A critical review of indoor chemical emissions and exposure led to the proposal of a modular mechanistic framework for predicting human exposure to SVOCs. The framework allows for integration of new knowledge, other chemical classes of indoor pollutants, and mechanistic processes relevant to human exposure indoors.
