Metabolite elucidation of 2-fluoro-deschloroketamine (2F-DCK) using molecular networking across three complementary in vitro and in vivo models

Summary: This study investigated the metabolites of 2-fluoro-deschloroketamine (2F-DCK) using in vitro experimental approaches and molecular networking to compare results. Thirteen metabolites were produced in vitro with additional seven metabolites found in post-mortem samples. The HLMs and HepaRG cell models were complementary in identifying specific 2F-DCK metabolites, with nor-2F-DCK and a hydrogenated metabolite proposed as reliable markers for detection in HRMS libraries.

Summary: Methoxetamine (MXE), a dissociative substance, has been present on the designer drug market as a ketamine-substitute since 2010. Studies have shown that MXE affects brain processing, causing long-lasting behavioral abnormalities, neurotoxicity, and alterations in neurological, sensorimotor, and cardiorespiratory functions. Mechanistic evaluations indicate involvement of serotonin and 5-HT2 receptors, as well as NMDA and GABA receptors, in mediating the central effects of MXE.

The research revealed the stability of 2F-DCK and ketamine (KET) in wastewater, as well as their consumption levels in different cities, providing valuable real-time data for the government to develop new policies.
