ER-directed TREX1 limits cGAS activation at micronuclei

Lisa Mohr et al.

Summary: The study demonstrates that the endoplasmic reticulum (ER)-associated nuclease TREX1 degrades micronuclear DNA upon micronuclear envelope rupture, inhibiting cGAS activation. Mutations in TREX1 disrupt its localization to micronuclei, increase micronuclear DNA damage, and enhance cGAS activation.

Chromothripsis drives the evolution of gene amplification in cancer

Ofer Shoshani et al.

Summary: The study reveals that chromothripsis is a major driver of circular extrachromosomal DNA (ecDNA) amplification, mediated by poly(ADP-ribose) polymerases (PARP) and DNA-dependent protein kinase (DNA-PKcs). The structural evolution of ecDNAs further increases drug tolerance, leading to rapid acquisition of tolerance to altered growth conditions. This mechanism accelerates genomic DNA rearrangement and amplification into ecDNA, contributing to cancer therapy resistance.

Fully phased human genome assembly without parental data using single-cell strand sequencing and long reads

David Porubsky et al.

Summary: This study introduces a reference-free workflow for diploid de novo genome assembly, combining single-cell strand sequencing with continuous long-read or high-fidelity sequencing data. By employing this strategy, the researchers successfully generated completely phased de novo genome assemblies for each haplotype of an individual of Puerto Rican descent, demonstrating high accuracy and contiguity with low switch error rates. A comparison of Oxford Nanopore Technologies and Pacific Biosciences phased assemblies identified regions that are preferential sites of contig breaks, regardless of sequencing technology or phasing algorithms.

Spatially resolved single-cell genomics and transcriptomics by imaging

Xiaowei Zhuang

Summary: The recent development of genome-scale imaging allows for single-cell omics analysis in intact cells and tissues, enabling gene expression profiling, identification of cell types, and spatial mapping in complex tissues. The high spatial resolution of these approaches also allows for determining the spatial organizations of the genome and transcriptome inside cells, which are key regulatory mechanisms for gene expression.

Single-cell lineages reveal the rates, routes, and drivers of metastasis in cancer xenografts

Jeffrey J. Quinn et al.

Summary: This study used a Cas9-based lineage tracer to track the development and metastasis of lung cancer cells in a mouse model over months, revealing heterogeneity in metastatic capacity and genetic differences that drive invasiveness. The study also uncovered some genes driving metastasis and an unexpected suppressive role of KRT17 in metastasis.

RAD21 is a driver of chromosome 8 gain in Ewing sarcoma to mitigate replication stress

Xiaofeng A. Su et al.

Summary: This study investigates the reason for chromosome 8 gains in 50% of Ewing sarcomas, finding that trisomy 8 mitigates replication stress induced by EWS-FLI1 through gaining a copy of RAD21. RAD21 promotes tumorigenesis through single gene copy gain, potentially explaining some recurrent aneuploidies in cancer.

Structural variant evolution after telomere crisis

Sally M. Dewhurst et al.

Summary: Telomere crisis has been found to induce chromothripsis and breakage fusion bridge (BFB) cycles in vitro. This study shows that telomere crisis can generate a broader spectrum of structural variations, suggesting that cancers without chromothripsis and BFB cycles may have also originated from telomere crisis.

Gene copy-number changes and chromosomal instability induced by aneuploidy confer resistance to chemotherapy

Marica Rosaria Ippolito et al.

Summary: Mitotic errors causing aneuploidy can lead to genome instability, affecting gene copy number and influencing cancer cells' resistance to chemotherapy. This may explain why some chemotherapy treatments fail.

Chromosomal instability accelerates the evolution of resistance to anti-cancer therapies

Devon A. Lukow et al.

Summary: The study found that transient chromosomal instability can accelerate the development of resistance to anti-cancer therapies in tumor cells, as well as provide phenotypic plasticity for them to adapt to various stressful environments. Furthermore, aneuploidy may be an underexplored cause of therapy failure in human tumors.

Role of spatial patterns and kinetochore architecture in spindle morphogenesis

Pablo Lara-Gonzalez et al.

Summary: The mitotic spindle is a macromolecular machine responsible for faithful chromosome segregation during cell division. Assembly of the spindle is guided by the 'Search & Capture' principle in structured space. Nucleation of noncentrosomal microtubules near kinetochores and changes in kinetochore architecture promote rapid and accurate assembly of the spindle.
