Mitochondrial stress induced by continuous stimulation under hypoxia rapidly drives T cell exhaustion

Nicole E. Scharping et al.

Summary: Continuous TCR signaling and hypoxia increase Blimp-1, suppressing mitochondrial reprogramming and generating reactive oxygen species, leading to T cell exhaustion. Mitigating T cell-intrinsic ROS and reducing tumor hypoxia can limit T cell exhaustion, synergizing with immunotherapy.

A clinically applicable and scalable method to regenerate T-cells from iPSCs for off-the-shelf T-cell immunotherapy

Shoichi Iriguchi et al.

Summary: The study presents a feeder-free, scalable differentiation process of human induced pluripotent cells (iPSCs) into T-cells with T-cell receptor dependent anti-tumor function in vitro and in vivo, providing a platform for clinical application and research on human T-cell differentiation and biology.

TCR-engineered T cells targeting E7 for patients with metastatic HPV-associated epithelial cancers

Nisha B. Nagarsheth et al.

Summary: In a first-in-human phase 1 trial, T cells targeting HPV-16 E7 showed promising results in treating metastatic epithelial cancers, with objective clinical responses observed in 6 out of 12 patients. This suggests that genetically engineered T cell therapy may be effective in treating common carcinomas, but further studies are needed to understand the resistance mechanisms in advanced epithelial cancer.

Lifileucel, a Tumor-Infiltrating Lymphocyte Therapy, in Metastatic Melanoma

Amod A. Sarnaik et al.

Summary: This study showed that Lifileucel is effective in treating advanced melanoma, especially in patients who are refractory to PD-1 or PD-L1 therapy. The product provides durable responses and addresses a major unmet need in patients with limited treatment options after approved therapy.

Metabolic reprogramming of terminally exhausted CD8+ T cells by IL-10 enhances anti-tumor immunity

Yugang Guo et al.

Summary: The study demonstrates that a half-life-extended IL-10-Fc fusion protein can enhance the function of terminally exhausted CD8(+) tumor-infiltrating lymphocytes by promoting oxidative phosphorylation, leading to improved response to cancer immunotherapy.
