Progress on the role of extrachromosomal DNA in tumor pathogenesis and evolution

Summary: The amplification of oncogenes on extrachromosomal DNA (ecDNA) provides cancer cells with a new mechanism to adapt to the tumor microenvironment and accelerate tumor evolution. ecDNA lacks centromeres and has uneven inheritance, leading to increased tumor heterogeneity and competitive advantage for daughter cells with numerous copies of oncogenes. This non-Mendelian genetic mechanism promotes tumor invasiveness and drug resistance, contributing to poorer survival outcomes.
