Summary: In this study, the cytochrome P450 monooxygenase CYP107D1 was engineered for the stereo- and regioselective 7 beta-hydroxylation of lithocholic acid to yield ursodeoxycholic acid. By identifying specificity-determining residues and utilizing a synthetic small library, a nearly perfectly regio- and stereoselective triple mutant was developed, which produces UDCA at significantly higher levels than the initial variant. This biocatalyst offers new possibilities for environmentally friendly synthesis of UDCA from biological waste product.

Summary: In this study, the oxidizing activity of CYP109B1 was reconstituted by coupling redox pairs or fusing it to the reductase domain of two self-sufficient P450 enzymes. The best redox pair for CYP109B1 was found to be Fdr_0978/Fdx_1499 from Synechococcus elongatus, showing over 99% conversion with 73% 15 beta selectivity for testosterone. The enzyme displayed good activity and selectivity toward testosterone derivatives, with a shift in selectivity for specific substrates.

CyProduct is a novel machine learned in silico cytochrome P450 (CYP450) metabolism prediction suite that accurately predicts metabolic byproducts for a specified molecule and a human CYP450 isoform. The suite includes three modules for predicting reactions, bond sites, and generating metabolic byproducts, and outperformed other packages in predicting metabolites.
