Summary: This study established a type 2 diabetic mouse model of triple-negative mammary carcinoma and investigated the effect of metformin on tumor characteristics and immune/metabolic microenvironment. The results showed that diabetic mice had larger mammary tumors with altered stromal profile compared to nondiabetic/lean mice, and metformin treatment effectively normalized glucose levels and decreased metastatic seeding. These findings indicate the impact of diabetes and obesity on tumor cell attributes and metformin's ability to revert tumor cell and stromal changes in an obese and diabetic host environment.

O-GlcNAcylation Enhances NUSAP1 Stability and Promotes Bladder Cancer Aggressiveness

Summary: This study found that NUSAP1 is highly expressed in bladder cancer cells and tissues, and its up-regulation significantly promotes proliferation and inhibits apoptosis. Furthermore, the study revealed that O-GlcNAc expression is elevated in bladder cancer tissues and cells, and up-regulation of O-GlcNAc increases NUSAP1 protein expression and stability. Knockdown of OGT inhibits proliferation and tumorigenesis while promoting apoptosis in bladder cancer cells, and these effects are reversed when NUSAP1 is up-regulated.

O-GlcNAcylation of SRPK2 at a nuclear localization signal and subsequent nuclear import through binding to importin a reveals a general mechanism for protein nuclear import in cells. This work highlights the important role of O-GlcNAc in posttranscriptional regulation and suggests importin a as a reader of an O-GlcNAcylated nuclear localization signal.
