Summary: Deficiency of PGRN can reduce levels of human APP and A beta in young male mice, increase markers associated with microglial activation near A beta plaques, and up-regulate lysosome proteins with enhanced nuclear translocation of TFE3 leading to inflammation in response to A beta fibril treatment. The findings suggest a sex- and age-dependent effect of PGRN on APP metabolism and its role in regulating lysosomal activities and inflammation in plaque-associated microglia.
