Summary: Blocking the interaction between AIF and CypA using AIF fragment AIF(370-394) or its shorter variant AIF(381-389) is protective against glutamate-induced neuronal cell death and brain injury. The structural characterization of AIF(381-389) reveals a beta-hairpin conformation with reduced flexibility, leading to improved antiapoptotic activity and enhanced proteolytic stability compared to the parent peptide. This study provides insights for designing AIF/CypA inhibitors with better pharmacokinetic and pharmacodynamics features based on the 3D model of the AIF(381-389)/CypA complex obtained through NMR.
