A genome-scale CRISPR screen reveals factors regulating Wnt-dependent renewal of mouse gastric epithelial cells

Kazuhiro Murakami et al.

Summary: Safe utilization of the regenerative potential of adult stem cells for clinical applications requires a comprehensive understanding of the regulatory mechanisms. Epithelial organoid cultures accurately mimic in vivo stem cell-driven epithelial renewal, offering a great ex vivo platform for studying key regulatory mechanisms. Through genome-scale CRISPR knockout screening, Alk, Bclaf3, and Prkra were identified as potential regulators of gastric epithelial differentiation and could suppress stemness/proliferation.

Ultra-high-throughput single-cell RNA sequencing and perturbation screening with combinatorial fluidic indexing

Paul Datlinger et al.

Summary: The development of 'single-cell combinatorial fluidic indexing' (scifi) allows for cost-effective single-cell sequencing on a massive scale, combining preindexing of entire transcriptomes with single-cell RNA-seq using microfluidics. This approach significantly increases throughput and enables sample multiplexing in a straightforward manner.
