Pharmacological inactivation of the prion protein by targeting a folding intermediate

Giovanni Spagnolli et al.

Summary: The researchers introduced a new drug discovery approach called Pharmacological Protein Inactivation by Folding Intermediate Targeting (PPI-FIT), which targets folding intermediates of disease-causing proteins. Testing this approach on the cellular prion protein, they identified ligands that could stabilize a folding intermediate and promote its degradation, supporting the idea that modulating protein levels by acting on their folding pathways may have therapeutic potential. Moreover, recent computational advancements in simulating biochemical processes have allowed for investigating protein regulation mechanisms with realistic physics-based models at an atomistic level.

Neuroinflammation in Prion Disease

Bei Li et al.

Summary: Neuroinflammation, characterized by microglial activation and astrogliosis, is a common feature in neurodegenerative conditions like prion diseases. Microglia have a neuroprotective role, while reactive astrocytes can propagate prions and contribute to neurodegeneration. Targeting neuroinflammation could be a novel approach for treating prion diseases and understanding other neurodegenerative disorders.

Oral administration of repurposed drug targeting Cyp46A1 increases survival times of prion infected mice

Tahir Ali et al.

Summary: Prion diseases are fatal, infectious, and incurable neurodegenerative disorders caused by misfolding of the cellular prion protein. A brain-specific enzyme, Cyp46A1, plays a crucial role in eliminating brain cholesterol, which impacts prion conversion. Activating Cyp46A1 with the anti-retroviral medication EFV showed promising results in mitigating prion propagation and prolonging lifespan in mice, suggesting a potential therapeutic approach for prion diseases.

Genetic prion disease-related mutation E196K displays a novel amyloid fibril structure revealed by cryo-EM

Li-Qiang Wang et al.

Summary: This study revealed the structure of amyloid fibrils formed by full-length human PrP with E196K mutation, which differs from wild-type PrP and hamster brain-derived prion fibrils. The results highlight the importance of familial mutations in inducing different prion strains.
