Repurposing FDA approved drugs inhibiting mitochondrial function for targeting glioma-stem like cells

Sandipan Datta et al.

Summary: GBM tumors contain glioma stem-like cells (GSCs) and differentiated GBM cells (d-GCs) with differences in mitochondrial metabolism. Inhibiting mitochondrial function selectively targets GSCs and may offer a potential therapeutic option for GBM treatment. Further studies are needed to explore the effectiveness of mitochondrial inhibitors as a targeted therapy for GBM.

Modulation of Nogo receptor I expression orchestrates myelin-associated infiltration of glioblastoma

Jun-Hee Hong et al.

Summary: The study identified NgR1 as a key player in suppressing myelin-associated infiltration in glioblastoma cells through deubiquitinating activity of USP1. Pimozide upregulates NgR1 by post-translational targeting of USP1, sensitizing glioma stem cells to myelin inhibition. Downregulation of NgR1 expression is associated with highly infiltrative characteristics and poor survival in primary human glioblastoma.

Calmodulin/CaMKII-γ mediates prosurvival capability in apicidin-persistent hepatocellular carcinoma cells via ERK1/2/CREB/c-fos signaling pathway

Wei-Chung Hsu et al.

Summary: This study revealed that CaM/CaMKs enhance the survival capability of liver cancer cells exposed to apicidin by activating signaling cascades, promoting tumor growth and inhibiting apoptosis. Inhibition of CaM effectively attenuated these effects, providing a promising target for cancer treatment.

Chlorpromazine induces cytotoxic autophagy in glioblastoma cells via endoplasmic reticulum stress and unfolded protein response

Silvia Matteoni et al.

Summary: The study demonstrates that chlorpromazine (CPZ) hinders malignant features of glioblastoma (GBM) cells by perturbing endoplasmic reticulum (ER) stress and inducing an intense autophagic response. This cytotoxic effect is selective for GBM cells while being cytoprotective for normal cells, suggesting a potential novel treatment approach for GBM patients.

Anticancer Properties of the Antipsychotic Drug Chlorpromazine and Its Synergism With Temozolomide in Restraining Human Glioblastoma Proliferation In Vitro

Silvia Matteoni et al.

Summary: Chlorpromazine, a medication used for psychiatric disorders for decades, has shown potential in treating glioblastoma by inhibiting cell viability, reducing cloning efficiency, decreasing stemness gene expression, and synergizing with temozolomide. The promising results have led researchers to initiate a Phase II clinical trial to evaluate the efficacy of adding chlorpromazine to the standard treatment protocol for GBM patients.
