Dissecting the immune landscape of tumor draining lymph nodes in melanoma with high-plex spatially resolved protein detection

Georgia M. Beasley et al.

Summary: In melanoma patients, the infiltration of CD3 cells in the sentinel lymph nodes varies among patients, with higher infiltration of CD11c and CD3 cells in the SLN tumor region associated with longer overall survival. Expression of DC activation markers such as CD86, HLA-DR, and OX40L on CD11c cells within the SLN tumor region correlates with patient survival, with lowest expression in patients with shorter survival and highest in patients with longer survival. Further studies are needed in a larger cohort to validate these findings.

Lung Histopathology in Coronavirus Disease 2019 as Compared With Severe Acute Respiratory Sydrome and H1N1 Influenza A Systematic Review

Lida P. Hariri et al.

Summary: The histopathologic pattern of COVID-19, H1N1 influenza, and SARS patients predominantly involves acute-phase diffuse alveolar damage (DAD), with more frequent pulmonary microthrombi observed in COVID-19 and SARS patients compared to H1N1 influenza. Further research is needed to validate these findings and explore their clinical implications.

B cells and cancer: To B or not to B?

Wolf Herman Fridman et al.

Summary: Recent studies have suggested that B cells in the tumor microenvironment may play a significant role in presenting tumor-associated antigens to T cells and producing antibodies that enhance antigen presentation, ultimately impacting cancer clinical outcomes. Immune complexes formed by B cells can also have deleterious effects through inflammation, angiogenesis, and immunosuppression via macrophage and complement activation.

Fecal microbiota transplant overcomes resistance to anti-PD-1 therapy in melanoma patients

Diwakar Davar et al.

Summary: The study demonstrated that fecal microbiota transplantation combined with anti-PD-1 therapy can overcome resistance to anti-PD-1 in a subset of PD-1 refractory melanoma patients, leading to clinical benefits. This approach induced changes in the gut microbiome and reprogrammed the tumor microenvironment, ultimately enhancing the efficacy of anti-PD-1 treatment.

Deciphering the state of immune silence in fatal COVID-19 patients

Pierre Bost et al.

Summary: COVID-19 patients show unique immune features, including lung accumulation of naive lymphoid cells, systemic expansion and activation of myeloid cells, and immune suppression. Loss of monocyte-driven and neutrophil-driven immune suppression may lead to fatal outcomes in severe patients. On the other hand, a lung CXCR6(+) effector memory T cell subset may be associated with better prognosis in severe COVID-19 patients.

Lisa M. McGinnis et al.

Summary: Novel multiplexing technologies for tissues have greatly impacted basic and translational research by allowing highly multiplexed immunohistochemistry or in situ RNA measurements with objective biomarker quantification and increased sensitivity. These technologies offer unprecedented insights into tissue biology and biomarker science, but implementation requires extensive time and resources.

The pathogenic role of epithelial and endothelial cells in early-phase COVID-19 pneumonia: victims and partners in crime

Marco Chilosi et al.

Summary: The understanding of the complex pathogenesis of COVID-19 interstitial pneumonia has been discussed in this article based on biopsies carried out in early/moderate phases and histology data obtained at postmortem analysis. The observations from autopsies cannot fully explain some clinical, radiological and physiopathological features observed in SARS-CoV-2 interstitial pneumonia, suggesting the need for further research to understand the discrepancies.

Covid-19 Interstitial Pneumonia: Histological and Immunohistochemical Features on Cryobiopsies

Claudio Doglioni et al.

Summary: In this study, lung samples from 12 patients with moderate Covid-19 pneumonia were analyzed via transbronchial lung cryobiopsy, revealing early pathological changes such as acute lung injury spots and alveolar type II cell hyperplasia. Additionally, unique endothelial and inflammatory cell phenotypes were observed, providing insights into the pathogenesis of Covid-19 interstitial pneumonia.

Broad betacoronavirus neutralization by a stem helix-specific human antibody

Dora Pinto et al.

Summary: The study identified five monoclonal antibodies from COVID-19 convalescent individuals that cross-react with multiple betacoronavirus spike glycoproteins, with one antibody (S2P6) showing broad neutralization of viruses from three different subgenera. This antibody reduced viral burden in hamsters challenged with SARS-CoV-2 by inhibiting membrane fusion, providing a framework for the design of pan-betacoronavirus vaccines for broad protection.

Risk assessment and seroprevalence of SARS-CoV-2 infection in healthcare workers of COVID-19 and non-COVID-19 hospitals in Southern Switzerland

Luca Piccoli et al.

Summary: The study found that up to 10% of HCWs in acute care hospitals in Southern Switzerland have detectable SARS-CoV-2 antibodies. HCWs with exposure to COVID-19 patients have only a slightly higher absolute risk of seropositivity compared to those without, suggesting that the use of PPE and other measures aiming at reducing nosocomial viral transmission are effective. Households contact with known COVID-19 cases represents the highest risk of seropositivity.

Functional characterization of CD4+ T cell receptors crossreactive for SARS-CoV-2 and endemic coronaviruses

Arbor G. Dykema et al.

Summary: Our study confirms the presence of unique memory CD4(+) T cell clones recognizing both SARS-CoV-2 and CCCs. The lower avidity of crossreactive TCRs for SARS-CoV-2 may result in reduced proliferation capacity and impact clinical outcomes in COVID-19 patients. Further research is needed to fully understand the implications of these crossreactive T cell responses.
