A multicenter, randomized phase III trial of hetrombopag: a novel thrombopoietin receptor agonist for the treatment of immune thrombocytopenia

Summary: Hetrombopag, a novel thrombopoietin receptor agonist, has been shown to be efficacious in increasing platelet counts and reducing bleeding risks in adults with immune thrombocytopenia (ITP) in Phase I studies. In a Phase III study, hetrombopag demonstrated significantly higher response rates after 8 weeks of treatment compared to placebo, as well as maintained durable platelet responses throughout 24 weeks. The most common adverse events with 24-week hetrombopag treatment were upper respiratory tract infection, urinary tract infection, immune thrombocytopenic purpura, and hematuria.

In preclinical studies, it has demonstrated improvement in clinical scores and joint pathology in arthritis models, as well as inhibitory effects on activation and inflammatory activities of immune cells without inducing cell death.

Hetrombopag: First Approval

Summary: Hetrombopag, an oral nonpeptide thrombopoietin receptor agonist developed by Jiangsu Hengrui Pharmaceutical, received its first approval in China on June 16, 2021, as a second-line treatment for primary immune thrombocytopenia (ITP) and severe aplastic anaemia (SAA) in adults. The drug is also undergoing phase III development in China for chemotherapy-induced thrombocytopenia treatment. This article highlights the milestones in the development of hetrombopag that led to its first approval for ITP and SAA.

Mycophenolate Mofetil for First-Line Treatment of Immune Thrombocytopenia

Summary: In a multicenter, open-label, randomized trial on patients with immune thrombocytopenia, 91.5% of patients who received mycophenolate mofetil plus glucocorticoids had platelet levels above 100,000 per microliter, compared to 63.9% of patients who received glucocorticoids only. Quality-of-life measures of physical function and fatigue were worse in the mycophenolate mofetil group.

Sialylated immunoglobulin G: a promising diagnostic and therapeutic strategy for autoimmune diseases

Summary: Human immunoglobulin G (IgG), especially sialylated IgG, plays a crucial role in the diagnosis and management of autoimmune diseases by exerting significant anti-inflammatory effects. Current research indicates that IgG Fc N-glycosylations are more reliable diagnostic and monitoring biomarkers than total IgG autoantibodies. IgG sialylation-based therapies show promise in attenuating pathogenic autoimmunity in preclinical and clinical studies.
