Circulating Tumor DNA Markers for Early Progression on Fulvestrant With or Without Palbociclib in ER plus Advanced Breast Cancer

Ben O'Leary et al.

Summary: The study found that high ctDNA fraction, TP53 mutation, and FGFR1 amplification were associated with worse progression-free survival in patients with breast cancer receiving combination endocrine and CDK4/6 inhibitor therapy. These patients may benefit from inclusion in future trials of escalating treatment.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Prognostic Significance of Blood-Based Multi-cancer Detection in Plasma Cell-Free DNA

Xiaoji Chen et al.

Summary: The study evaluated the prognostic significance of cancer detection by the MCED test using longitudinal follow-up data, finding that cancers detected by the test had lower overall survival rates and could differentiate more aggressive cancers. Cancers not detected by the test had better prognosis, and the MCED test results were predictive of their outcomes.

The mutational landscape of human somatic and germline cells

Luiza Moore et al.

Summary: Mutations accumulate in normal human cells over a lifetime. This study compared mutational landscapes in 29 cell types from the soma and germline, revealing similarities and differences in the maintenance of these cell types. Spermatogonia, the stem cells generating sperm and origins of most genetic variation in humans, showed the lowest mutation rate potentially due to low rates of mutational processes and cell division.

Clonal Architecture of EGFR Mutation Predicts the Efficacy of EGFR-Tyrosine Kinase Inhibitors in Advanced NSCLC: A Prospective Multicenter Study (NCT03059641)

Xinghao Ai et al.

Summary: This study investigates clonal architecture in advanced non-small cell lung cancer (NSCLC), finding that EGFR clonal architecture serves as an independent prognostic indicator of the efficacy of EGFR-tyrosine kinase inhibitors (TKIs). Paired tissue and ctDNA can be analyzed for clonal architecture in advanced cancer.

Towards systematic nomenclature for cell-free DNA

Abel J. Bronkhorst et al.

Summary: The report classifies different types of cfDNA molecules in the human body based on origin, genetic traits, and locality, suggesting new terms and abbreviations to address the lack of universal cfDNA nomenclature. Recommendations for proper usage of these terms are made, focusing on oncology, prenatal testing, and post-transplant surgery surveillance in particular.
