Microglia and lipids: how metabolism controls brain innate immunity

Bruno Chausse et al.

Summary: Microglia are sensors of alterations in CNS physiology and undergo metabolic reprogramming to adapt to inflammatory responses. Lipid metabolism in activated microglia plays a crucial role in controlling functions such as migration, phagocytosis, and inflammatory signaling, with disturbances in these processes associated with brain function changes in neuroinflammatory disorders.

Protective role of microglial HO-1 blockade in aging: Implication of iron metabolism

Cristina Fernandez-Mendivil et al.

Summary: The study revealed that overexpression of HO-1 in microglial cells in aged mice leads to neurotoxic iron accumulation, oxidative stress, and cognitive decline. Inhibiting HO-1 or using an iron chelator can prevent these issues, providing new insights for treating age-related neurodegenerative diseases.

Microglial phagocytosis of neurons in neurodegeneration, and its regulation

Claire A. Butler et al.

Summary: Recent evidence suggests that excessive microglial phagocytosis of live neurons and synapses contributes to neurodegeneration in animal models of various brain diseases. Factors regulating microglial phagocytosis of neurons could be potential treatment targets to prevent neurodegeneration induced by excessive microglial phagocytosis of live neurons and synapses.

Dystrophic microglia are associated with neurodegenerative disease and not healthy aging in the human brain

Ryan K. Shahidehpour et al.

Summary: The study found an age-associated increase in dystrophic microglia in the hippocampus and frontal cortex, but this increase was proportional to the total number of microglia, suggesting aging alone does not explain the presence of dystrophic microglia. Dystrophic microglia, rather than hypertrophic microglia, were found to be the disease-associated microglia morphology. Furthermore, changes in iron homeostasis in dystrophic microglia were identified as a potential molecular mechanism driving the degeneration of microglia in neurodegenerative disease.

Age-dependent and region-specific alteration of parvalbumin neurons, perineuronal nets and microglia in the mouse prefrontal cortex and hippocampus following obesogenic diet consumption

Amy C. Reichelt et al.

Summary: The study reveals that excessive consumption of HFHS diets negatively impacts hippocampal and PFC function, resulting in reduced PV neurons and PNNs in the hippocampus of adult mice on HFHS diet, correlated with obesity, while no effects were seen in the PFC. Additionally, HFHS diet led to increased microglia in adults and activated microglia phenotypes in adolescents.

12-Hydroxyeicosapentaenoic acid inhibits foam cell formation and ameliorates high-fat diet-induced pathology of atherosclerosis in mice

Takahiro Nagatake et al.

Summary: This study investigated the impact of dietary intake of omega-3 fatty acids on atherosclerosis development and found that supplementing high-fat diet with alpha-linolenic acid-rich linseed oil improved atherosclerosis while inhibiting foam cell formation. The study also revealed a significant increase in the anti-inflammatory metabolite 12-hydroxyeicosapentaenoic acid (12-HEPE) in mice fed with linseed oil, which improved pulsatility and resistive indexes and inhibited macrophage foam cell transformation in a PPARγ-dependent manner.

A large-scale nanoscopy and biochemistry analysis of postsynaptic dendritic spines

Martin S. Helm et al.

Summary: Dendritic spines can be classified into mushroom-like and stubby shapes, which exhibit similar average protein copy numbers and topologies but differ in their correlation to synaptic strength. Stubby spines are less likely to respond adequately to dynamic changes in synaptic transmission compared to mushroom spines, possibly leading to their disappearance during brain maturation. This study provides a molecular view of dendritic spines through electron microscopy, quantitative biochemistry, super-resolution microscopy, and 3D molecular visualizations.

Probiotic Supplementation and High-Intensity Interval Training Modify Anxiety-Like Behaviors and Corticosterone in High-Fat Diet-Induced Obesity Mice

Parisa Foroozan et al.

Summary: In high-fat diet-induced obesity mice, probiotic supplementation and high-intensity interval training (HIIT) can reduce obesity, anxiety-like behaviors, and corticosterone levels. The combination of HIIT and probiotics may have a greater effect on anxiety indices compared to using them separately.
