Investigating the genetic architecture of noncognitive skills using GWAS-by-subtraction

Perline A. Demange et al.

Summary: Genomic structural equation modeling was used to estimate the genetic component of educational attainment independent of cognitive ability, finding that 57% of genetic variation in educational attainment is attributed to noncognitive skills. The study identified significant loci and a polygenic architecture contributing to educational attainment, with noncognitive genetics showing associations with personality traits, behavior, and psychiatric disorders. Noncognitive and cognitive-performance genetics were found to be similarly associated with socioeconomic success and longevity.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

The use of negative control outcomes in Mendelian randomization to detect potential population stratification

Eleanor Sanderson et al.

Summary: Mendelian randomization analysis assumes no association between genetic variants and outcomes other than through the exposure of interest, but population stratification can introduce confounding. Negative control outcomes are suggested for detecting potential confounding in MR studies. Regular use of negative control outcomes is recommended to detect population stratification in MR studies.

Cross-cancer evaluation of polygenic risk scores for 16 cancer types in two large cohorts

Rebecca E. Graff et al.

Summary: This study investigated polygenic risk score (PRS)-specific pleiotropy across 16 cancers in European ancestry individuals, finding positive and inverse cross-cancer associations. The study identified bidirectional relationships between certain cancer types and highlighted the potential for informing risk prediction and precision cancer prevention strategies.
