Summary: As biological therapeutics continue to develop, chemical methods for modifying recombinant peptides and proteins are becoming more valuable. Recent advancements have been made in modifying specific amino acids, but achieving regioselectivity in the presence of chemically identical residues remains challenging. This mini-review explores the latest progress in regioselective chemical modification of peptides and proteins using microenvironment and/or proximity-driven effects for achieving selectivity.

Boosted activity by engineering the enzyme microenvironment in cascade reaction: A molecular understanding

Summary: The study demonstrates that modulating enzyme microenvironment can significantly enhance enzyme activity, leading to increased catalytic efficiency by adjusting the pH-activity profiles of model enzymes. Modified enzymes with extended operational pH range exhibit higher activity. Spectroscopy and simulation studies reveal that the activity enhancement is due to a favorable microenvironment supporting the preservation of enzyme native structures under external perturbations.

To overcome this, researchers have been devoted to expanding the protein sequence space through chemical modifications and/or incorporation of noncanonical amino acids.

Efficiently Enantioselective Hydrogenation Photosynthesis of (R)-1-[3,5-Bis(trifluoromethyl)phenyl] ethanol over a CLEs-TiO2 Bioinorganic Hybrid Materials

Summary: By utilizing a biohybrid photocatalytic system based on artificial cross-linked enzymes (CLEs)-TiO2-Cp*Rh(bpy) photoenzyme, a successful synthesis of chiral alcohol intermediates is achieved, with hydrogen and electrons provided by water and photocatalytic systems reducing NADP+ to NADPH. The photoenzyme-catalyzed combined reductive synthesis has a yield of 41.2% and a very high enantiomeric excess value (>99.99%), showcasing excellent reusability and environmental friendliness.
