A CRISPR-driven colorimetric code platform for highly accurate telomerase activity assay

Summary: The Telomeric Repeat Amplification Protocol (TRAP) is commonly used for assessing telomerase activity, but often faces challenges such as false-negative results due to inhibiting factors in cell lysates or tissue samples. A new colorimetric code system based on CRISPR-Cas12a and AuNPs probe has been developed to rapidly analyze telomeric repeat DNA in TRAP products, enabling easy detection of telomerase activity and false-negatives. This platform has shown promise in detecting telomerase activity in clinical liver cancer samples with high sensitivity and specificity. Furthermore, a Cas9-mediated lateral flow assay has been demonstrated as a convenient and accurate method for telomerase activity assay on a single test strip.
