Antivirals that target the host IMPα/β1-virus interface

Alexander J. Martin et al.

Summary: Small molecule inhibitors targeting the IMP alpha/beta 1-dependent nuclear import have shown robust antiviral activity, especially against viruses like HIV-1, DENV, and ZIKV. Compounds such as ivermectin directly target this interface and have shown broad-spectrum activity against various viruses, including SARS-CoV-2. Rigorous RCTs are underway to evaluate the therapeutic potential of targeting the IMP alpha/beta 1-virus interface for antiviral development.

An in-silico study on selected organosulfur compounds as potential drugs for SARS-CoV-2 infection via binding multiple drug targets

Liya Thurakkal et al.

Summary: The shift towards multi-targeted small molecules has opened up innovative pathways in drug discovery, which was utilized in investigating drugs for COVID-19. Seventy-six organosulfur compounds were screened for their interactions with multiple SARS-CoV-2 target proteins through molecular dynamics simulation, with Lurasidone and its derivatives showing significant binding affinity against five proteins. Pharmacokinetics, ADMET properties, and target prediction studies further supported the efficacy against SARS-CoV-2.

A systematic review of experimental evidence for antiviral effects of ivermectin and an in silico analysis of ivermectin's possible mode of action against SARS-CoV-2

Robert T. Kinobe et al.

Summary: Ivermectin has shown potential as an antiviral agent by inhibiting viral replication in vitro, including various virus families. The drug may target nuclear location signals in the SARS-CoV-2 genome, potentially blocking viral proteins from reaching a virus-resistant state. While initial studies show promising results, further research is needed to validate the efficacy of ivermectin in clinical settings.
