Summary: A nanoporous polyethylene glycol hydrogel with self-cleaning capacity was developed for direct amplification of nucleic acid in complex matrices without sample pretreatment. The hydrogel facilitated nucleic acid amplification by adsorbing and removing inhibitors during isothermal amplification, resulting in successful amplification even in the presence of inhibitors. Digital quantification of pathogens in whole blood was achieved within 20 minutes, demonstrating high specificity and low detection limit.
