Autophagy regulation by acetylation-implications for neurodegenerative diseases

Sung Min Son et al.

Summary: Acetylation plays a crucial role in neurodegenerative diseases by affecting the clearance and aggregation of proteins, leading to neuron loss and impaired function. Therapeutic targeting of acetylation to restore healthy autophagic activity may have neuroprotective effects in various neurodegenerative diseases. These findings highlight the importance of understanding the role of acetylation in regulating autophagy for the development of potential treatments for neurodegenerative disorders.

Autophagy inhibition attenuates TGF-β2-induced epithelial-mesenchymal transition in lens epithelial cells

Yan Sun et al.

Summary: This study demonstrated that TGF-beta 2 induces autophagy flux during EMT progression of LECs, with autophagy activation enhancing fibrogenic responses and cell migration, while autophagy inhibition alleviates EMT markers and cell migration. Additionally, modulation of Smad signaling by autophagy was observed, and ATG7-silenced LECs showed antifibrosis effect induced by TGF-beta 2. Overall, intervention/inhibition of autophagy could attenuate TGF-beta 2-induced EMT in LECs, providing insights for preventing and treating fibrotic cataracts and other fibrotic diseases.

YAP plays a crucial role in the development of cardiomyopathy in lysosomal storage diseases

Shohei Ikeda et al.

Summary: Mutations in lysosomal genes lead to lysosomal dysfunction, resulting in lysosomal storage disorder (LSD) and causing functional abnormalities in major organs. In LSD, autophagy is inhibited and accumulation of autophagosomes occurs, with YAP playing a key role in mediating cardiomyopathy.

YAP promotes autophagy and progression of gliomas via upregulating HMGB1

Min Zhao et al.

Summary: YAP over-expression enhances glioma cell autophagy and progression by promoting the translocation of HMGB1 from nucleus to cytoplasm, suggesting a potential therapeutic target in GBM. The combination of chemo-radiotherapy with pharmacological autophagy inhibition for treating GBM patients with YAP high expression may be a promising clinical opportunity.

Autophagy-dependent survival is controlled with a unique regulatory network upon various cellular stress events

Orsolya Kapuy et al.

Summary: Autophagy serves as a type of programmed cell death but also plays a crucial role in cell survival under acceptable levels of stress. Through a controlled network, including components like autophagy inducer, autophagy controller, mTORC1, and autophagy executor, the switch-like characteristic of autophagy induction is achieved, allowing for specific activation in response to cellular stress. This control network, with components that act as molecular switches, ensures a robust response in various stress processes, preventing permanent hyperactivation of autophagy.

α-Catenin levels determine direction of YAP/TAZ response to autophagy perturbation

Mariana Pavel et al.

Summary: This study reveals that autophagy can have opposing effects on the regulation of YAP/TAZ, depending on cellular context, due to a negative feedback loop involving alpha-catenin.

Cell type-specific YAP1-WWTR1/TAZ transcriptional responses after autophagy perturbations are determined by levels of α-catenins (CTNNA1 and CTNNA3)

Mariana Pavel et al.

Summary: The YAP1-WWTR1/TAZ transcription co-factors play a crucial role in cell growth and may be dysregulated in cancer. The study reveals that alpha-catenin levels act as a switch in determining the response of YAP1-WWTR1/TAZ signaling after autophagy perturbations, with a complex feedback loop between YAP1-WWTR1/TAZ and autophagy. The integration of experimental and theoretical data provides insights into cell-specific and time-dependent responses to autophagy manipulations, potentially relevant in various cancer contexts.

Guidelines for the use and interpretation of assays for monitoring autophagy (4th edition)

Daniel J. Klionsky et al.

Summary: The guidelines presented in this article aim to assist researchers in selecting and interpreting methods to examine autophagy, as well as to help reviewers provide constructive critiques of reports focused on these processes. It emphasizes that no single assay is perfect for every situation, and multiple techniques may be needed to properly monitor autophagy in different experimental settings. The article also highlights the importance of targeting multiple autophagy-related genes in genetic approaches to block autophagy effectively.
